Tissue analysis is essential for understanding the distribution, metabolism, and potential toxicity of therapeutic oligonucleotides. However, extracting oligonucleotides from complex matrices such as liver remains a significant analytical challenge. 👩‍🔬

Proteins, lipids, endogenous nucleic acids, nucleases, and other matrix components can reduce extraction efficiency and compromise the reproducibility of LC-MS analysis. Effective sample preparation is therefore critical to isolate the analyte, minimize matrix interferences, and achieve reliable quantitation.

In this application note, we demonstrate a workflow using Biotage® Oligo SPE for the extraction of mipomersen from liver tissue, achieving recoveries of ≥70% with reliable quantitation.
